which date will consider if i vary my wife application ( PBS dep) for ilr

my wife pbs dep visa is going to expire 27-aug-2018 i am going to apply her extension as ( pbs dep)
on 24 aug 2018 she is only short of 32 Days for ILR means she can apply her ilr on 28 sept 2018 .

Her history is

visa stamped date is : 24- oct-2013
entry in uk : 31-dec-2014 in between she got her extension as well
pbs expire date is 27-aug 2018

as per new rule ( PBS dep tier 1 gen) only fill application online
i have a plan to fill online application for her extension to cover the 32 days
after that on 28 sep -2018 i will go to peo and vary my application

Q : so please can any body tell me which date will be consider if i vary my wife application

Is the date when i submit my online application pbs tier 1 general dep

or

Is the date when i go to peo on 28-sep -2018 ?

After submit (pbs depnt tier1 gen) online application how many days after we can give bio metric : ?

the main reason is why i am more concern to safe the one application money ( PBS extension )

any suggestion will be appreciated

and is there any proper format or letter to vary the application as well
