Hi, I currently want to apply for citizenship online and I never have biometric card, I only have ILR on my old passport. Do I need to get the biometric enrolment from HO before I submitted the application

You do not need to get a Biometric card first, ILR on your old passport is fine. You do however have to enroll your biometrics (finger prints and digital photo) after you have submitted the online form once you have made your appointment at UKCVAS Sopra Steria. They are two separate things completely.
