1. What is "relevant period". E.g. if I have PR do I just submit evidence for 5 years?
relevant a period is up to 10 years working back from the date of the AN application - not to be confused with PR.
2. Will 1 letter from employer suffice, if I had worked for this one employer constantly for the past 6 years?
YES - I had 1 letter for 10 years!

I think you might be wrong about the 10 years.
Guide, page 5, states: "Evidence of lawful residence during the 5 (or, if the applicant is married to or in civil
partnership to a British citizen, 3) years before the date of the application"
That what my friend has confirmed as well
I think 10 years of evidence of NI only applies if Home Office has a reason to mistrust you, as Guide page 7 states.
Also, the new online application does not even mention NI contributions.
